چکیده

The biosynthesis of 14 protein bands from 50S ribosomes was studied during the outgrowth of Bacillus subtilis W23 spores. The time at which the synthesis of each protein band begins and the patterns of synthesis that each displays were determined by comparing the incorporation of (3)H-phenylalanine during five intervals throughout the first hour of outgrowth with the incorporation of (14)C-phenylalanine between 145 and 150 min. The majority of the proteins in the 14 bands analyzed were synthesized as early as 5 to 10 min. The bands could be grouped on the basis of their labeling patterns as well as the time required for them to achieve a high rate of synthesis. Three bands did not achieve a high rate of synthesis until after 30 min of outgrowth, and at least one of the proteins from these bands appeared to be located on the exterior of the 50S subunit. The significance of the proteins from these three bands is discussed in terms of the late formation of new, mature 50S subunits during outgrowth.

Properties of Bacillus megaterium and Bacillus subtilis mutants which lack the protease that degrades small, acid-soluble proteins during spore germination.

During germination of spores of Bacillus species the degradation of the spore's pool of small, acid-soluble proteins (SASP) is initiated by a protease termed GPR, the product of the gpr gene. Bacillus megaterium and B. subtilis mutants with an inactivated gpr gene grew, sporulated, and triggered spore germination as did gpr+ strains. However, SASP degradation was very slow during germination of...

Interaction of apurinic/apyrimidinic endonucleases Nfo and ExoA with the DNA integrity scanning protein DisA in the processing of oxidative DNA damage during Bacillus subtilis spore outgrowth.

Oxidative stress-induced damage, including 8-oxo-guanine and apurinic/apyrimidinic (AP) DNA lesions, were detected in dormant and outgrowing Bacillus subtilis spores lacking the AP endonucleases Nfo and ExoA. Spores of the Δnfo exoA strain exhibited slightly slowed germination and greatly slowed outgrowth that drastically slowed the spores' return to vegetative growth. A null mutation in the di...

Genetic analysis of the developmental processes during germination and outgrowth of Bacillus subtilis spores with temperature-sensitive mutants.

HE developmental processes in bacteriophage T4 have been studied extenTsively with temperature-sensitive mutants and amber mutants (EDGAR, DENHARDT and EPSTEIN 1964; EDGAR and LIELAUSIS 1964). In Bacillus subtilis, the sporulation process has been studied with asporogenous mutants ( RYTER, SCHAEFFER and IONESCO 1966; TAKAHASHI 1965). We examined the developmental processes involved in the trans...

How moist heat kills spores of Bacillus subtilis.

Populations of Bacillus subtilis spores in which 90 to 99.9% of the spores had been killed by moist heat gave only two fractions on equilibrium density gradient centrifugation: a fraction comprised of less dense spores that had lost their dipicolinic acid (DPA), undergone significant protein denaturation, and were all dead and a fraction with the same higher density as that of unheated spores. ...

Relationship between protein and ribonucleic acid synthesis during outgrowth of spores of Bacillus cereus.

The rate of protein and ribonucleic acid (RNA) synthesis was examined during the outgrowth of spores of Bacillus cereus T in a chemically defined medium. RNA synthesis started 2.5 min after the initiation of germination, and protein synthesis after 4 min. Addition of a complete amino acid supplement and uracil supported high rates of RNA and protein synthesis throughout outgrowth. To determine ...
